Product Specific Information

Description:
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits (gamma, delta, epsilon) are essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular pathways leading to activation and proliferation.

• Functional activity (in vitro stimulation)

Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Storage and Handling:
Use in a sterile environment.

Target Information

The CD3 complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. These subunits have long cytoplasmic tails that associate with signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum. It is not expressed on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
