Description:
The HIT3a monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with the other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR with HIT3a initiates an intracellular biochemical pathway resulting in cellular activation and proliferation.

Applications Reported:
The HIT3a antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Use in a sterile environment.

0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. Activation through TCR involves CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related immunoglobulin superfamily members encoded on chromosome 11. CD3 subunits have cytoplasmic tails associated with signal transduction molecules via tyrosine-based motifs. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation. It is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Reduced T lymphocytes may be observed in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
