• Antibody is tested in direct ELISA.
• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD73 (Ecto-5′-nucleotidase, 5′-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
It consists of a dimer of two identical 70 kDa subunits linked to the plasma membrane via a glycosyl phosphatidylinositol anchor. CD73 serves as a lymphocyte differentiation marker and its deficiency is associated with various immunodeficiency diseases.
Other 5′ nucleotidase forms in cytoplasm and lysosomes differ by substrate affinity and ion requirements. The CD73 gene is localized to chromosome 6q14–q21.
Defects in CD73 can lead to joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
