Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD73 (Ecto-5′-nucleotidase, 5′-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
CD73 is a dimer of two identical 70 kDa subunits attached to the plasma membrane via a glycosyl phosphatidylinositol linkage. It serves as a marker for lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
Other forms of 5′-nucleotidase exist in the cytoplasm and lysosomes, distinguishable by substrate affinity, magnesium ion requirement, ATP activation, and phosphate inhibition.
The CD73 gene is localized to chromosome 6q14–q21. Defects in this gene can cause joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms of CD73 have been identified.
