Target Information
The SMAD1 protein belongs to the SMAD family, homologous to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.
SMAD1 mediates bone morphogenetic protein (BMP) signaling, which regulates cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, SMAD1 is phosphorylated and activated by BMP receptor kinase, forming a complex with SMAD4 for transcriptional regulation. SMAD1 is also targeted by SMAD-specific E3 ubiquitin ligases (SMURF1, SMURF2) for ubiquitination and proteasomal degradation.
Alternatively spliced transcript variants encoding the same protein have been observed.
