Target Information
AXL is a chronic myelogenous leukemia-associated oncogene also linked to colon cancer and melanoma. The AXL protein is a receptor tyrosine kinase that transduces extracellular signals into the cytoplasm by binding growth factors such as vitamin K-dependent GAS6. It plays roles in cell proliferation and apoptotic cell clearance.
TAM receptors (TYRO3, AXL, MERTK) are important for phagocytosis of apoptotic cells. Mice lacking all three TAM receptors show degenerative phenotypes and autoimmune symptoms similar to systemic lupus erythematosus.
Axl may act as an entry receptor for filoviruses and serves as a marker for a subset of human dendritic cells (Siglec6+/Axl+). Its expression can be upregulated by TLR ligands (e.g., LPS, poly I:C). Soluble Axl, generated by proteolytic cleavage, may be elevated in inflammatory or cancer conditions.
