Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription through coactivator or corepressor recruitment.
There are two main forms, ER alpha and ER beta, encoded by ESR1 and ESR2 genes, respectively. Alternative RNA splicing produces at least four ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and play essential roles in sexual development, reproductive function, and bone metabolism. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
