Target Information

LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of a family of membrane glycoproteins. LAMP2 provides selectins with carbohydrate ligands and may play a role in tumor cell metastasis, as well as in protection, maintenance, and adhesion of the lysosome.

Prior to posttranslational modification, LAMP2 is approximately a 46 kDa polypeptide. The mature, functional form is extensively glycosylated with N-linked and O-linked oligosaccharides, resulting in a total molecular weight of about 100–130 kDa.

LAMP2 consists of a large amino-terminal intra-lysosomal domain, a hydrophobic transmembrane domain, and a short carboxyl-terminal cytoplasmic tail. It is used to monitor chaperone-mediated autophagy, a selective form of autophagy.

There are three variant forms of LAMP2: LAMP-2A, LAMP-2B, and LAMP-2C. Together with CD107a/LAMP-1, LAMP2 is a major constituent of the lysosomal membrane. The LAMP proteins are involved in lysosome biogenesis and are required for fusion of lysosomes with phagosomes. LAMP2 is an important regulator of successful phagosomal maturation.

Deficiency of LAMP2 leads to accumulation of autophagosomes in many tissues, causing cardiomyopathy and myopathy (Danon’s disease).
