Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with other CD3 subunits (gamma and delta), is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.
This antibody is validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.5 µg) per test.
A test is defined as staining a cell sample (10⁵–10⁸ cells/test) in 100 µL final volume.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on human chromosome 11. These subunits associate with cytoplasmic signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells. It is absent on B and NK cells. Altered CD3 expression can be observed in autoimmune diseases. Defects in the CD3 gene are linked to CD3 immunodeficiency.
