Product Specific Information

The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular signaling resulting in activation and proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes (≤0.5 µg/test). Recommended titration for optimal assay performance.
Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-57).
Excitation: 488–561 nm; Emission: 775 nm; Laser: Blue, Green, Yellow-Green.

Target Information

The CD3 complex plays a crucial role in transducing antigen-recognition signals into T cells and regulating TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, members of the immunoglobulin superfamily encoded on chromosome 11. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
