Product Specific Information

Description:
The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by the majority of thymocytes, a subpopulation of mature T-helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from RPA-T4 and does not cross-block binding.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL total volume). Suitable for 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD4 antigen is involved in MHC class II recognition and acts as a co-receptor for HIV. It is primarily expressed on T-helper lymphocytes but also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain regions of brain and gut. CD4 functions in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in infectious or immune-related CNS diseases. Multiple spliced transcripts have been identified (RefSeq, July 2017).
