Target Information

PER2 (Period circadian protein homolog 2) is a mainly nuclear protein that undergoes nucleocytoplasmic shuttling through interactions with other circadian oscillator proteins or by phosphorylation. It belongs to the basic helix-loop-helix family of transcription factors and contains a PAC (PAS-associated C-terminal) domain and two PAS (PER-ARNT-SIM) domains.

PER2 is a key component of the circadian core oscillator complex, which includes CRY proteins, CLOCK or NPAS2, BMAL1 or BMAL2, CSNK1D and/or CSNK1E, TIMELESS, and other PER proteins. It plays a critical role in generating circadian rhythms and acts as a negative regulator in the circadian transcriptional loop by interacting with regulatory proteins and transporting them to the nucleus.

PER2 is widely expressed, showing strong expression in skeletal muscle and pancreas, and slight expression in lung. Mutations in PER2 are associated with familial advanced sleep-phase syndrome (FASPS).
