Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription via coactivator or corepressor recruitment.

There are two forms of estrogen receptors—alpha and beta—encoded by separate genes (ESR1 and ESR2, respectively). Alternative RNA splicing produces at least four isoforms of estrogen receptor alpha (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and essential for sexual development, reproductive function, and bone physiology. They are implicated in pathological processes such as breast cancer, endometrial cancer, and osteoporosis.
