Target Information
Calcium-permeable channels such as TRPV6 participate in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger.
Depending on the tissue, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA).
TRPV6 acts as a calcium-selective cation channel likely involved in Ca²⁺ uptake in various tissues, including intestinal reabsorption. The channel is activated by low internal calcium levels and exhibits inward rectification.
Inactivation occurs through a rapid Ca²⁺-dependent and a slower Ca²⁺-calmodulin-dependent mechanism, possibly regulated by phosphorylation.
In vitro, TRPV6 is slowly inhibited by Mg²⁺ in a voltage-independent manner. Heteromeric assembly with TRPV5 modifies channel properties, and TRPV5–TRPV6 heteromultimers show voltage-dependent gating.
TRPV6 interacts with TRPV5, S100A10, and the ANAX2–S100A10 heterotetramer, which is required for plasma membrane trafficking. It also interacts with BSPRY and calmodulin.
