Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms (345 and 395 kDa) are expressed in dividing cells. Ki-67 is detectable during active cell cycle phases (G1, S, G2, mitosis) but absent in resting cells (G0).
During interphase, expression is nuclear; during mitosis, Ki-67 associates with chromosomes and heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 serves as a marker to determine the fraction of proliferating cells.

This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using human peripheral blood cells with Foxp3/Transcription Factor Buffers Set (Product #00-5523-00).

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin via its FHA domain. Expression pattern shifts with cell cycle stage:

  • G1: perinucleolar expression
  • G2: nuclear matrix localization
  • M phase: chromosomal association

Ki-67 is widely used as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis. It is useful for determining cell growth fraction in neoplasms and correlates strongly with tumor grade. Ki-67 is also used as a neuronal marker of cell cycling and proliferation.
