Product Specific Information

The 3D12 monoclonal antibody reacts with human CCR7 (EBI-1, CD197), a G-protein-coupled chemokine receptor functioning as a receptor for CCL19 and CCL21. CCR7 expression distinguishes naive, central, and effector memory T cells and plays a role in migration of memory T cells to inflamed tissues. It is also expressed on dendritic cells (DCs) and increases during DC maturation, affecting chemotaxis, architecture, endocytosis, and survival.

• Applications Tested: Pre-titrated and validated on human peripheral blood cells
  • Recommended use: 5 µL (0.6 µg) per test (100 µL final volume)
  • Cell number: 10⁵–10⁸ cells/test
  • Incubation: ≥45 min at 2–8°C for optimal staining

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CCR7 is a G protein-coupled receptor induced by Epstein-Barr virus (EBV), mediating effects on B lymphocytes. It is expressed in blood, bone marrow, lymph node, and intestine, predominantly in lymphoid tissues and activated B/T lymphocytes. CCR7 regulates memory T-cell migration and dendritic cell maturation. Ligand: CCL19/ECL. Related receptors include CCR1–CCR5 and Duffy antigen, involved in T-cell chemotaxis and phagocyte migration.
