Product Specific Information

The IP26 monoclonal antibody reacts with the alpha/beta chain of human TCR, which is expressed by the majority of peripheral T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of human peripheral blood cells at 5 µL (0.2 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.
Staining can be obstructed when OKT3 (anti-human CD3) antibody is used as a co-stain.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes.

Buffer Information:
This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Use 5 µL per sample (regardless of the number of NovaFluors in your panel) for optimal staining.
For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures that allow engineered fluorescent signatures with reduced spillover and spread.

Target Information

The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer non-covalently associated with invariant CD3 subunits (gamma, delta, eta, zeta), all required for surface expression.
TCR engagement induces T cell activation, differentiation, and cytokine production.
Most T cells express TCR alpha/beta and either CD4 or CD8 (single positive). A small subset lacks both (double negative).
Gamma/delta T cells are mainly found in epithelia and show distinct antigen recognition patterns.
Alterations in TCR expression or recognition are linked to autoimmune diseases and tumor immunity.
Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and lupus, suggesting compensatory immune mechanisms.
