Product Specific Information

The 3D12 monoclonal antibody reacts with human CCR7 (CD197), a G-protein-coupled chemokine receptor that binds CCL19 and CCL21. CCR7 is expressed on T cells and dendritic cells (DCs), and plays a role in distinguishing naïve, central, and effector memory T cell populations. It is involved in T cell migration and DC maturation, chemotaxis, and survival.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated for normal human peripheral blood cells. Use 5 µL (0.6 µg) per test in 100 µL final volume.
  • Recommended incubation: ≥45 minutes at 2–8°C for optimal staining.
  • Included reagents: 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered nucleic acid dye structures designed to reduce spillover and spread.

Target Information

CCR7 is a G-protein-coupled receptor induced by Epstein-Barr virus (EBV) and expressed in blood, bone marrow, lymph nodes, and intestinal tissues. It regulates migration of memory T cells to inflamed tissues and promotes dendritic cell maturation. Ligands include chemokines CCL19 and CCL21. CCR family members (CCR1–CCR5) are key in T cell chemotaxis and phagocyte migration to inflammatory sites.
