Product Specific Information

The 3D12 monoclonal antibody reacts with human CCR7 (CD197), a G-protein-coupled chemokine receptor that binds CCL19 and CCL21. CCR7 is expressed on T cells and dendritic cells (DCs), and plays a role in T cell migration, DC maturation, and chemotaxis toward lymph nodes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.56 µg) per test.

• Recommended incubation: ≥45 minutes at 2–8°C
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer
• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining
• Add 5 µL of CellBlox Plus per sample before combining with cells
• For single-color controls: 5 µL buffer per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CCR7 is a member of the G protein-coupled receptor family and a subfamily of chemokine receptors. It is induced by Epstein-Barr virus (EBV) and mediates its effects on B lymphocytes. CCR7 is expressed in blood, bone marrow, lymph node, and intestine, particularly in activated B and T lymphocytes. It regulates memory T cell migration to inflamed tissues and promotes dendritic cell maturation. Ligands include CCL19 (ECL) and CCL21.
