Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low but play a critical role in allergic responses by binding to mast cells and basophils through Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume).

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) compared to single-color stained cells. Set compensation with cells when possible. Similar effects observed with AbC Total Antibody Compensation beads (Product # A10497).

Light Sensitivity: Protect from light; dye is sensitive to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark, with minimal impact on brightness and FRET efficiency.

Target Information

IgE is one of five mammalian immunoglobulin classes. Composed of two heavy (epsilon) and two light chains, IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylactic shock) via Fc receptor binding on basophils and mast cells, inducing degranulation and allergic responses. It also contributes to parasite immunity.
