Product Specific Information

The ISA-3 monoclonal antibody reacts with human ICOS (Inducible COStimulatory molecule), also known as H4, CRP-1, and AILIM. ICOS is a T cell-specific activation molecule and a member of the CD28/CTLA-4 family. It plays a key role in T cell activation and humoral immune responses, including memory B cell and plasma cell generation.

ICOS binds to its ligand B7h/B7RP-1 on activated antigen-presenting cells (APCs) and inflamed peripheral tissues. Plate-bound ISA-3 induces production of IL-4, IL-5, IL-10, and other cytokines, but not IL-2. ISA-3 was generated against human ICOS antigen.

Clone C398.4A (anti-mouse ICOS/H4) cross-reacts with human ICOS but binds a different epitope. For brightest staining of ICOS on activated human T cells, use Product #13-9948-82 or Product #12-9948-42.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry at 5 µL (0.5 µg)/test.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter. Ensure the instrument can detect this fluorochrome.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize non-specific interactions.

Compensation values for Super Bright 780-conjugated antibodies may differ between microsphere beads and stained cells; compensation with cells is recommended.

Light sensitivity: Protect from light to prevent photo-oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
