Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except erythrocytes and platelets. The cytoplasmic portion has tyrosine phosphatase activity essential for lymphocyte activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter. Ensure instrument compatibility.

Compensation Notes:
Higher compensation values may occur in violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) or AbC Total Antibody Compensation beads (Product # A10497). Set compensation with cells when possible.

Staining Panel Recommendation:
For multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, constituting about 10% of lymphocyte surface proteins. Absent in non-hematopoietic tissues, CD45 regulates Src-family kinases and is crucial for lymphocyte development and antigen signaling. Multiple isoforms arise via alternative splicing, differing in extracellular domains. CD45 influences integrin-mediated signaling, apoptosis, and cytokine receptor regulation.
