Product Specific Information

The MIH5 monoclonal antibody reacts with mouse B7-H1 (PD-L1). B7-H1 is a member of the B7 family (~40 kDa) and belongs to the Ig superfamily. It is expressed on T, B, NK, and dendritic cells, acting as a ligand for PD-1. Interaction of PD-1 with PD-L1 or PD-L2 results in inhibition of T and B cell responses. MIH5 is reported to be a blocking antibody.

• Applications Reported: Suitable for flow cytometric analysis.
• Applications Tested: Validated for flow cytometric analysis of mouse splenocytes at ≤0.06 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
  Optimize antibody titration for best results.

Super Bright 780 Characteristics:

• Excited by violet laser (405 nm), emits at 780 nm.
• Recommended filter: 780/60 bandpass or equivalent.
• Ensure instrument compatibility for detection.

Usage Notes:

• When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Compensation values may vary when using UltraComp eBeads ([Product # 01-2222-42]) or AbC Total Antibody Compensation beads ([Product # A10497]); set compensation using cells when possible.
• Sensitive to photo-induced oxidation; protect from light.
• Fixed samples can be stored up to 3 days at 4°C using IC Fixation Buffer ([Product # 00-8222]) or 1-step Fix/Lyse Solution ([Product # 00-5333]).

Target Information

Programmed death receptor ligand 1 (PD-L1, B7-H1) is a B7 family member that binds PD-1 receptor, negatively regulating TCR signaling. PD-L1 reduces T-cell proliferation and cytokine production, and is highly expressed on many murine and human cancers, up-regulated by IFN-γ stimulation. PD-L1 plays a role in tumor immune evasion.
