Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low and play a key role in allergic responses by binding to mast cells and basophils through the Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils, existing in both soluble and membrane-bound forms, and IgE21 recognizes both.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor Technology:
Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

IgE is one of five mammalian immunoglobulin classes. It consists of two heavy (epsilon) chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylactic shock) by binding to Fc receptors on basophils and mast cells, triggering degranulation and allergic responses. It also contributes to parasite immunity.
