Target Information
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans. EGFR belongs to the HER/ERbB family of receptor tyrosine kinases, including ERbB2, ERbB3, and ERbB4. Ligand binding (e.g., EGF, TGF-α) induces receptor dimerization and autophosphorylation, activating downstream signaling pathways such as Ras-ERK, PI3K-Akt, Jak-STAT, and PKC. These cascades regulate cell growth and proliferation. EGFR phosphorylation at Y1086 promotes GRB2 binding and MAPK pathway activation. Receptor activation leads to endocytosis and degradation or recycling. EGFR mutations and overexpression are linked to various cancers, including lung, head and neck, brain, and breast tumors, and are often associated with poor prognosis.
