Product Specific Information

The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and mediates interactions with B cells via CD72, playing a key role in T-B cell communication and proliferation. The UCHT2 antibody recognizes primate CD5.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤5 µL (0.4 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize dilution for best performance.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence spectra, minimizing spillover and spread.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein expressed on mature T lymphocytes, most thymocytes, and certain B cell subsets. It regulates T-cell activation and proliferation. CD5 is found on:

• 95% of thymocytes
• 72% of peripheral blood lymphocytes

In lymph nodes, CD5 reactivity is localized in T cell areas. It is expressed in T cell leukemias, lymphomas, and activated T cells.
Diseases associated with CD5 dysfunction include thymus cancer and Richter’s Syndrome.
