Product Specific Information

The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and mediates T-B cell interactions via CD72 binding.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (0.4 µg)/test. A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume. Cell count may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring nucleic acid dye structures designed for engineered fluorescence with minimized spillover and spread.

Target Information

CD5 is a 67 kDa human T-lymphocyte transmembrane glycoprotein expressed on most thymocytes, mature T cells, and subsets of B cells. It regulates T-cell proliferation and is found on ~95% of thymocytes and ~72% of peripheral blood lymphocytes. CD5 expression is also observed in T-cell leukemias, lymphomas, and activated T cells. Associated diseases include thymus cancer and Richter’s Syndrome.
