Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays a key role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/CD8β) and functions as a co-receptor with TCR for MHC-I molecules. CD8α is essential for MHC-I binding and is also found as homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
