Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase)는 핵 내 세포사멸(apoptosis)을 조절하는 기질 단백질로서, caspase-3에 의해 활성화되는 DNase의 억제제 역할을 합니다. ICAD는 두 가지 isoform을 가지며, 45kDa의 ICAD-L(DFF45)과 35kDa의 ICAD-S(DFF35)가 존재합니다. ICAD-L만이 기능적으로 활성화되어 CAD를 억제하며, caspase-3에 의해 절단되어 CAD가 DNA 절편화를 수행할 수 있도록 합니다. 또한 ICAD는 CAD의 접힘을 돕는 chaperone 역할을 하며, 최근에는 ICAD가 대장암 및 뇌암 세포에서 정상적인 세포사멸 신호 전달 경로를 회복시키는 잠재적 표적으로 보고되고 있습니다. ICAD 유전자는 두 가지 상이한 isoform을 암호화하는 대체 스플라이싱 전사체를 가집니다.
