Target Information
FAS (Fas, CD95, APO-1)는 46 kDa의 막관통형 당단백질로, TNFR (tumor necrosis factor receptor) 수퍼패밀리의 세포사멸 수용체로 작용합니다.
FAS는 약 26 kDa의 가용성 형태로도 존재하며, 자극 시 FAS-associated death domain (FADD)을 통해 death-inducing signaling complex (DISC)를 형성하고, caspase 활성화를 유도하여 세포사멸(apoptosis)을 일으킵니다.
또한 FAS는 NF-κB, MAPK3/ERK1, MAPK8/JNK 신호경로를 활성화하며, 정상 섬유아세포 및 T세포의 증식 신호 전달에도 관여합니다.
여덟 가지 이상의 스플라이싱 변이체가 보고되었으며, 막관통 도메인이 결여된 형태는 전체 길이 단백질에 의한 세포사멸을 음성 조절할 수 있습니다.
Fas/Fas ligand 시스템은 AIDS, 간염, 암 등 다양한 질환과 관련이 있으며, 항바이러스 면역 반응에서 중요한 역할을 하는 것으로 알려져 있습니다.
