Product Specific Information

This antibody detects endogenous levels of Histone H2A.X only when phosphorylated at Tyr143.

Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. In eukaryotes, DNA double-strand breaks (DSBs) induce phosphorylation of serine 139 at the C-terminus of H2AX, resulting in gamma-H2AX. This phosphorylation can be detected by Western blotting or immunofluorescence to assess DSB frequency. While phosphatidylinositol 3-kinases are implicated in H2AX phosphorylation, the exact kinase—ATM, DNA-PK, or ATR—remains under investigation.

H2A.X consists of 143 amino acids and is synthesized during both G1 and S phases. Its mRNA exists in two forms: a 1600-base polyA+ form and a 575-base replication-type polyA− form. The gene maps to chromosome 11q23.2–q23.3 in humans. H2A.X contributes to nucleosome assembly and DNA structure, and specifically mediates the interaction of MDC1, a DNA repair protein, at sites of DNA damage.
