Product Specific Information

The LG.7F9 monoclonal antibody reacts with mouse CD27, a lymphocyte-specific member of the TNFR superfamily.
CD27 is expressed by virtually all mature T cells and a subpopulation of B cells, mainly memory B cells. In mouse, CD27 is found on nearly all thymocytes excluding CD46-CD8- precursors.
CD27 binds to CD70 and plays an important role in T cell–B cell interaction and effector cell maturation. LG.7F9 cross-reacts with human and rat CD27.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test. A test is defined as staining a cell sample in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Super Bright 436 can be excited with a violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

Staining Panel Guidance:
Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright dyes to minimize non-specific polymer interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily (includes CD40 and CD30). These receptors regulate cell proliferation and death.
CD27 and its ligand CD70 are predominantly expressed in lymphocytes. CD27 is a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subset of B cells.
Activation of T cells via TCR-CD3 complex upregulates CD27 and releases soluble CD27 (sCD27, 28–32 kDa), detectable in plasma, urine, or spinal fluid. sCD27 serves as a prognostic marker for acute and chronic B cell malignancies.
RgpA, a cysteine proteinase, degrades CD27 and counteracts its activation. CD27-binding protein (SIVA) binds to CD27 and induces apoptosis.
Diseases associated with CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
