Target Information

Tuberous sclerosis complex (TSC) is an autosomal dominant tumor syndrome caused by mutations in either the TSC1 or TSC2 tumor suppressor genes. The protein products form a complex that negatively regulates the mammalian Target of Rapamycin (TOR) pathway, influencing cell growth and cycle progression.
TSC1/TSC2 complex stimulates Rheb GTPase activity, thereby reducing TOR activation. Overexpression of TSC1 and TSC2 decreases the GTP/GDP ratio bound to Rheb and inhibits cell growth.
A shorter 40 kDa isoform of TSC1 exists, though its function remains unknown.
