Product Specific Information
The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed on B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation, induces Ig isotype-switching, and prevents apoptosis in combination with IL-4. The CD40–CD154 interaction is crucial for T-B cell communication and immune regulation.
5C3 is commonly used for B cell activation in in vitro assays.

Target Information
CD40 is a single-chain glycoprotein of the TNF/NGF receptor superfamily, showing homology to CD30. It is expressed on all B cells (except plasma cells), some epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 is essential for immune and inflammatory responses, including:
- T cell-dependent immunoglobulin class switching
- Memory B cell development
- Germinal center formation
The transcription factor AKNA regulates CD40 expression, influencing cell interactions. CD40 interaction with TNFR2 mediates signal transduction. The CD40–CD154 pathway is implicated in amyloid-β-induced microglial activation, suggesting a role in Alzheimer’s disease. Two alternatively spliced transcript variants of CD40 are known.
