Product Specific Information

The WM59 monoclonal antibody reacts with human CD31 (PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 plays a role in cell adhesion, cell–cell and cell–matrix interactions, and signal transduction.

Applications Reported:
This antibody is validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm). Recommended filter: 710/50 BP (695/40 BP acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Clone-specific performance after fixation should be verified empirically.

Excitation / Emission / Laser:
488 nm / 710 nm / Blue Laser

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor that regulates T and B cell signaling via ITIM motifs, providing docking sites for protein-tyrosine phosphatases upon phosphorylation. It is mainly located at endothelial cell junctions and mediates homophilic binding important for cell–cell interactions. CD31 functions in integrin-mediated adhesion, transendothelial migration, angiogenesis, apoptosis, immune regulation, and thrombosis. It is a key regulatory molecule in the vascular system.
