Product Specific Information
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two genes:
- CD16A (FcγRIIIA): expressed in macrophages and NK cells as a transmembrane form
- CD16B (FcγRIIIB): expressed in neutrophils as a GPI-anchored form
Two polymorphisms (NA-1 and NA-2) exist for CD16B. Individuals homozygous for NA-2 show lower phagocytic capacity than NA-1. CD16 binds IgG immune complexes, showing preferential binding to IgG1 and IgG3 isotypes. Upon IgG binding, both isoforms trigger signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.06 µg) per test.
A test is defined as the amount of antibody staining cells in a final 100 µL volume. Recommended cell number: 10⁵–10⁸ cells/test.
Fluorochrome Information:
- Excitation: 405 nm
- Emission: 436 nm
- Recommended filter: 450/50 bandpass or equivalent
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Super Bright Polymer Dyes are sold under license from Becton, Dickinson and Company.
Target Information
CD16 encodes a receptor for the Fc portion of IgG, involved in immune complex clearance, antibody-dependent cellular cytotoxicity (ADCC), and modulation of viral infection susceptibility.
The FCGR3A gene (CD16A) is expressed on NK cells as a transmembrane glycoprotein, while FCGR3B (CD16B) is expressed on neutrophils with a phosphatidylinositol linkage.
Mutations in FCGR3A are associated with immunodeficiency 20 and increased susceptibility to herpes zoster and systemic lupus erythematosus.
Alternative splicing produces multiple isoforms.
