Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (low-affinity Fc gamma RIII). CD16 exists as two isoforms: FcγRIIIA (on monocytes, macrophages, NK cells) and FcγRIIIB (on neutrophils). Expression patterns vary among primates.

Binding of IgG activates signal transduction pathways leading to antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, cytokine release, and antigen presentation.

• Recognizes the same epitope as CB16; distinct from B73.1 clone.
• Reported for flow cytometric analysis.
• Tested at 5 µL (0.06 µg) per test (100 µL final volume).
• Recommended cell number: 10⁵–10⁸ cells/test.
• Excitation: 405 nm; Emission: 436 nm (use 450/50 bandpass filter).
• Compatible with violet laser.
• Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright conjugates to minimize non-specific interactions.

Target Information

CD16 encodes a receptor that binds the Fc portion of IgG and mediates immune complex clearance, ADCC, and other immune functions. The gene FCGR3A is similar to FCGR3B, both located on chromosome 1.

• FCGR3B: Expressed on neutrophils, anchored via phosphatidylinositol linkage
• Associated disorders: Immunodeficiency 20, Herpes Zoster, recurrent viral infections, systemic lupus erythematosus, alloimmune neonatal neutropenia
• Alternative splicing produces multiple isoforms
