Target Information

Calcium-permeable channels such as TRPV6 are involved in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA). TRPV6 functions as a calcium-selective cation channel and participates in calcium uptake in various tissues, including intestinal calcium reabsorption.

The channel is activated by low internal calcium levels and shows inward rectification. Inactivation occurs via rapid Ca(2+)-dependent and slower Ca(2+)-calmodulin-dependent mechanisms, potentially regulated by phosphorylation. TRPV6 is slowly inhibited by Mg(2+) in a voltage-independent manner in vitro. It forms heteromeric assemblies with TRPV5, modifying channel properties and exhibiting voltage-dependent gating. TRPV6 interacts with TRPV5, S100A10, ANAX2-S100A10 heterotetramer, BSPRY, and calmodulin; the interaction with S100A10 is required for plasma membrane trafficking.
