Target Information

Calcium-permeable channels such as TRPV6 are involved in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Depending on the tissue, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA).
TRPV6 functions as a calcium-selective cation channel, likely participating in Ca²⁺ uptake and reabsorption in various tissues including the intestine. The channel is activated by low internal calcium levels and exhibits inward rectification.
Inactivation mechanisms include rapid Ca²⁺-dependent and slower Ca²⁺-calmodulin-dependent processes, potentially regulated by phosphorylation. In vitro, TRPV6 is slowly inhibited by Mg²⁺ in a voltage-independent manner. Heteromeric assembly with TRPV5 modifies channel properties, and TRPV5–TRPV6 heteromultimers show voltage-dependent gating.
TRPV6 interacts with TRPV5, S100A10, ANAX2–S100A10 heterotetramer, BSPRY, and calmodulin. The interaction with S100A10 is essential for trafficking to the plasma membrane.
