Product Specific Information

Description:
The G14D9 monoclonal antibody reacts with human Glycoprotein A Repetitions Predominant (GARP, also known as LRRC32 or Garpin). GARP is an ~80 kDa glycoprotein expressed on the cell surface. Expression has been reported in placenta, lung, kidney, heart, ovary, liver, skeletal muscle, and pancreas, as well as on megakaryocytes, platelets, and activated regulatory T (Treg) cells. GARP expression on activated Treg cells is essential for their suppressive function, possibly as a receptor for LAP/TGF-β.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometric analysis of stimulated normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test in a final volume of 100 µL. Recommended cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

GARP (Leucine Rich Repeat Containing 32, LRRC32) is a type I membrane protein with 22 leucine-rich repeats and a short intracytoplasmic region. Identified through Treg and Th cell gene expression profiling, it is expressed in CD4⁺, CD25⁺, FoxP3⁺ regulatory T cells and serves as a marker for Treg cells.
