• Recommended positive controls: human blood-derived granulocytes
• Neutralization Dose₅₀ (ND₅₀) for anti-human IL5RA is approximately 1–3 µg/mL in the presence of 0.5 ng/mL recombinant human IL-5 using the human TF-1 cell line.
• ND₅₀ indicates the antibody concentration required to achieve half-maximal inhibition of IL5RA-mediated IL5 response on responsive cells.
• The exact neutralization concentration depends on IL5 concentration and IL5RA expression level on the cell surface.

Target Information

Interleukin 5 (IL-5) is a soluble T cell-derived factor, also known as T cell replacing factor (TRF), that induces B cell and eosinophil growth and differentiation.
IL-5 acts through the IL-5 receptor (IL-5R), composed of two chains:

• Alpha chain (IL5RA): binds IL-5 with low affinity
• Beta chain: forms a high-affinity receptor complex with the alpha chain

The cytoplasmic domains of both subunits are essential for signal transduction. The membrane-proximal proline-rich sequence of IL5RA is critical for IL-5–induced proliferation, proto-oncogene expression, and tyrosine phosphorylation of proteins such as JAK1 and JAK2.
