Product Specific Information

The monoclonal antibody miap301 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein with a small cytoplasmic domain involved in adhesion through interactions with SIRP and non-covalently associated with β3 integrins (CD51/CD61, CD41/CD61). It also serves as a receptor for thrombospondin.

• No cross-reactivity is observed against human CD47.
• Expression is found in most hematopoietic cells (T cells, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic cells.
• Reported applications include Flow Cytometry, Immunoprecipitation, Western blot (nonreducing conditions), and Immunohistology of frozen tissue sections.

Tested Application Example:

• Flow Cytometric analysis of mouse splenocytes.
• Recommended ≤ 0.5 µg/test (100 µL final volume).
• Cell number range: 10⁵–10⁸ cells/test.
• Antibody titration recommended for optimal assay performance.

Target Information

CD47 (Integrin-Associated Protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and regulates their functions. Engagement of CD47 modulates signaling pathways such as activation of G proteins and intracellular calcium increase during cell adhesion. CD47 is a receptor for thrombospondin and plays roles in membrane transport and signal transduction.

• Broad tissue distribution; reduced on Rh erythrocytes.
• Acts as a ligand for CD172a (SIRPα), inhibiting macrophage phagocytosis of CD47-positive cells.
• Involved in cell migration, B cell adhesion, T cell activation, chondrocyte mechanical response modulation, and neuronal development—especially abundant in synapse-rich regions of brain and retina.
