Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Applications Reported:
Flow cytometric analysis, immunoprecipitation, immunohistochemical staining, immunoblotting (non-reducing conditions), and complement-dependent cytotoxicity.
For functional assays, use Functional Grade purified IM7 (Product #16-0441).

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells and splenocytes. Recommended ≤0.125 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize concentration for specific assay conditions.

Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein expressed on human leukocytes, brain white matter, and epithelial cells of intestine and breast. Multiple isoforms exist, with CD44H being predominant in normal tissues. It acts as a receptor for hyaluronic acid (HA) and is involved in cell-cell interactions, adhesion, and migration. CD44 participates in lymphocyte activation, recirculation, and homing, and may be upregulated in carcinomas with potential links to metastasis. Expression is found in hematopoietic, non-hematopoietic, and epithelial cells, as well as filopodia in cultured keratinocytes. Alternative splicing produces diverse isoforms. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus.
