Product Specific Information
Description:
The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated upon stimulation via BCR, CD40, or cytokine receptors. CD86 interacts with CD28 and CTLA-4 on T cells, playing a key role in T-B cell communication, T cell costimulation, and immune regulation.
Applications Reported:
This antibody is validated for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test (empirically determined).
Light Sensitivity:
PE-Cyanine5 is sensitive to photo-induced oxidation. Protect from light.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
CD86 is a ligand for CTLA-4 and CD28, acting as a costimulatory molecule in T-cell activation. Insufficient co-stimulation can induce tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed on antigen-presenting cells. It interacts with CD28 to activate T-cells and with CTLA-4 to inhibit activation. Alternative splicing produces multiple transcript variants. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
