Product Specific Information

Description:
The DX2 monoclonal antibody reacts with human CD95 (Fas, Apo-1), a 40–50 kDa member of the TNFR superfamily. CD95 is expressed on a wide range of hematopoietic and non-hematopoietic cells including monocytes, neutrophils, lymphocytes, and fibroblasts. Interaction of CD95 with its ligand (FasL) induces apoptosis and contributes to peripheral tolerance. DX2 does not block binding of EOS9.1, another antibody specific for human CD95. The DX2 monoclonal is also reported to recognize canine CD95.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number per test: 10⁵–10⁸ cells.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

FAS (Fas, CD95, APO-1):
A 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. Exists also in a soluble 26 kDa form. Upon stimulation, FAS aggregates and forms the death-inducing signaling complex (DISC), activating caspases and triggering apoptosis. It also activates NF-κB, MAPK3/ERK1, and MAPK8/JNK pathways, influencing proliferation and immune responses. Multiple isoforms exist; some lacking the transmembrane domain may inhibit apoptosis. The Fas/FasL system plays roles in diseases such as AIDS, hepatitis, and cancer, and is key in antiviral immune responses.
