This antibody was purified from whole rabbit serum prepared by repeated immunizations with the NFkB p65 peptide corresponding to the nuclear localization sequence (NLS) of the human protein conjugated to KLH using maleimide. A residue of cysteine was added to the amino terminal end to facilitate coupling.

Product Specific Information

Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

This affinity-purified antibody is directed against the NLS of human p65 and is useful in determining its presence in various assays. The epitope recognized overlaps the NLS of the p65 subunit of the NFkB heterodimer, allowing selective binding to the activated form of NFkB. Anti-NFkB p65 NLS may react non-specifically with other proteins.

Target Information

Nuclear factor kB p65 (NF-kB p65), encoded by the RELA gene on chromosome 11, is also known as RelA (v-rel avian reticuloendotheliosis viral oncogene homolog A) and is part of the Rel family of proteins. It forms heterodimers with p50 or p52 subunits of NF-kB. Upon TNF alpha binding to its receptor, IKK is phosphorylated, leading to phosphorylation and proteasomal degradation of IκB. NFkB p65 plays a key role in transcriptional regulation of immunoglobulin kappa gene in mature B-lymphoid cells.
