Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays an essential role in T cell development and function. RM4-5 binding is blocked by GK1.5.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test). Optimize antibody titration for best results.

Fluorochrome Information:
Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is designed for UV (355 nm) laser-equipped cytometers. Ensure instrument compatibility.

Buffer Recommendations:
Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) when combining multiple polymer dye-conjugated antibodies to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be fixed in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C (dark). Clone-specific performance should be verified empirically.

Target Information

The CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed on T helper cells and also found on monocytes, macrophages, dendritic cells, and in tissues such as thymus, lymph nodes, tonsils, and spleen. CD4 contributes to T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
