Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is lymphoma tissue.
Target Information
The INK4a locus on chromosome 9p21 is frequently affected in human tumors. It encodes two structurally distinct tumor suppressor proteins: p16INK4a and the alternative reading frame protein ARF (p14ARF in human and p19ARF in mouse).
These proteins interact with upstream tumor suppressors — retinoblastoma protein and p53 — and play key roles in cell cycle control and tumor suppression.
p14ARF can induce both G1 and G2 phase arrest in a p53-dependent manner. Differences between mouse and human ARF proteins may contribute to species-specific tumor susceptibility.
