Product Specific Information

The 104 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported:
Flow cytometric analysis, immunoprecipitation, immunohistochemical staining of frozen tissue sections.

Applications Tested:
Validated by flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimal titration recommended.

Target Information

CD45.2 (leukocyte common antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It plays a key role in lymphocyte development and antigen signaling by regulating Src-family kinases.

CD45.2 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains. Isoform expression varies by cell type and physiological state. In addition to immunoreceptor signaling, CD45.2 promotes cell survival via integrin-mediated pathways and contributes to DNA fragmentation during apoptosis. CD45RA is an isoform with restricted expression across lymphoid subtypes.
