Target Information
Voltage-gated potassium (Kv) channels are essential components of potassium currents in the heart and central nervous system. In the heart, Kv4 channels contribute to the repolarization phase of the action potential, while in the brain, they prevent reverse propagation of action potentials.
KChIPs (Kv channel interacting proteins) are calcium-binding proteins that associate with Kv4 channels and modulate their function. There are three known KChIPs:
- KChIP1: Expressed in the brain
- KChIP2: Expressed in brain, heart, and lung (with isoforms a, b, c)
- KChIP3 (DREAM/calsenilin): Expressed in brain and testis
KChIP3 not only modulates Kv4 currents but also acts as a DNA-binding transcriptional regulator by binding to the regulatory DRE and repressing transcription of early-response genes such as c-fos. It is also associated with presenilins and implicated in β-amyloid formation.
